Biotin-16-Aminoallyluridine-5'-Triphosphate
Description
Biotin-16-Aminoallyluridine-5'-Triphosphate (Biotin-16-UTP, Biotin-16-AA-UTP) can be incorporated via a number of RNA polymerases, including SP6-, T3- and T7-RNA polymerase to form labeled RNA. According to Fenn BJ, et al., T7 RNA polymerase does not significantly discriminate between the natural and the biotinylated nucleotide. Therefore, the number of biotinylated positions per transcript can be controlled by varying the ratio of the biotinylated NTP to the standard NTP. Biotinylated RNA probes are useful as a nonradioactive label in a number of applications including chromosome mapping, in situ hybridization and microarray hybridization. Biotin-16-U-modified RNA can be detected with a variety of streptavidin-based approaches.
